Thanks: interestingly each of the three methods is better in some parts of the image than others. However, Method C does seem to be the overall winner.

DOF lens correction factor

Is this unique for each lens, or would all lenses of the same type use the same correction factor? So, if I have a Nikon 105 macro lens, would all of them be the same? If yes, then maybe we could set up a list of the correction factor for most of the common lenses as a service to other users.

Best stacking method for protozoa

I am photography protozoa, which are basically transparent, and the resultant stacks are not as clear as I would like. Do any of you have experience with this, and do you haste any advice?

While I normally use Stackshot, I decided to try focus stacking, to compare the results. After setting up, I click choose Autofocus, then click on the nearest point on the screen image. The camera briefly focusses on the chosen spot, then goes completely out of focus. Maybe I am missing a key step, ...
