Is ist possible to shoot and process a panorama matrix in different focus planes and then generate a panorama with increased dof in ONE step ?

You will not be able to stitch different focus planes identically. This means that relative distances between objects will be different.
So I would suggest you to extend DOF first and then stitch panorama. Helicon Focus (win version) allows to process multiple stacks at once (menu\Add to queue\Add to queue multiple stacks ...)
