Can I use Helicon Focus with the pictures I've taken with my Sony Alpha A100 camera? Do they need to be shot in RAW then saved as tif files before using Helicon Focus for stacking?

Yes, you can open images taken by this cameras in Helicon Focus, both raw and JPEG.
